  • [Title] Fas and FasL expression on cells of two transplantable melanoma lines according to their different biological properties.
  • In tumours (e.g. melanoma), FasL expression possibly counteracts the Fas-positive effector T cells that infiltrate into tumours, and consequently the Fas/FasL interaction can contribute to the escape of tumour cells from the systemic immune response.
  • In this study we examined differences in Fas and FasL expression on cells from the hamster melanotic melanoma line (Ma) and a more aggressive amelanotic melanoma line (Ab).
  • We also tried to find out whether the Fas/FasL expression induces an ability to undergo spontaneous apoptosis in these two transplantable melanoma lines.
  • Isolated transplantable melanoma cells were incubated for 4 and 24 hours and after that time the expression of Fas and FasL was estimated by flow cytometry.
  • The results show that there was no Fas expression, although FasL was detected on both melanoma cell lines.
  • Therefore the data reported by other authors indicate that a lack or a low level of Fas expression and an ectopic expression of FasL on melanoma cells can be an escape mechanism of the tumour, to avoid host immune responses.
  • The content of FasL-positive melanotic melanoma cells was higher than in amelanotic melanoma cells and increased with the prolongation of the incubation time.
  • FasL expression on amelanotic melanoma cells was detected after 24 hours at a level similar to that on melanotic melanoma cells after 4 hours incubation time.
  • FasL expression on melanoma cells can induce apoptosis in cytotoxic T lymphocytes and NK cells which are responsible for tumour cells elimination.
  • The results obtained suggest that the Fas/FasL system does not play any significant role in spontaneous apoptosis of two melanoma cell lines.

  • BACKGROUND: Primary malignant melanoma of the vagina is extremely rare, accounting for 0.3-0.8% of all malignant melanomas.
  • True amelanotic vaginal melanoma showing no melanin on histological examination is exceedingly rare, accounting for only 2% of all vaginal melanomas.
  • CASE REPORT: We describe a 31-year-old female patient who presented with locally advanced amelanotic melanoma of the vagina, with no evidence of metastatic spread on the computerized tomography (CT) scan, but who was subsequently diagnosed as suffering from metastatic disease by positron emission tomography (PET)-CT performed a few weeks following posterior pelvic exenteration.
  • CONCLUSION: Specific immunohistochemical staining with melanoma markers should be performed to confirm or exclude a diagnosis of amelanotic melanoma in all patients presenting with a vaginal mass composed of undifferentiated epithelioid malignant cells.
  • Fluorodeoxyglucose (FDG)-PET-CT should be performed as part of the preoperative evaluation, to identify the presence or absence of metastatic disease in all patients with vaginal melanoma.

  • To identify genes differentially regulated in PML-1 melanoma cells, we performed DNA microarray and two-dimensional matrix-assisted laser desorption ionization-time of flight mass spectrometry analyses.

  • Ocular neoplasms occurring in XP in order of frequency are squamous cell carcinoma, basal cell carcinoma and melanoma.
  • Malignant melanomas occur at an early age in patients with XP.
  • We report a case of XP with massive orbital melanoma in an eight-year-old boy which is unique due to its amelanotic presentation confirmed histopathologically.

  • Histological examination showed a malignant nodular neoplasm with 2 distinct but intimately admixed components: a malignant melanoma with a spindle component and an unusual glandular component.
  • A single melanocytic marker (MITF) was expressed in both components, raising the possibility of dual differentiation in a single tumor, rather than the alternative considerations of a collision tumor or a reactive pseudoepitheliomatous hyperplasia with eccrine duct lumen formation within a melanoma.
  • This unusual tumor with both melanocytic and epithelial components may represent a true melanocarcinoma, which becomes a plausible consideration, in view of melanoma plasticity and recent experimental evidence and speculation about the role of stem cells in melanoma.

  • A total of 64 RCM features were scored retrospectively and blinded to diagnosis in a consecutive series of RCM sampled, clinically equivocal, macules of the face (n=81 LM, n=203 benign macules (BMs)).
  • A LM score of > or = 2 resulted in a sensitivity of 85% and specificity of 76% for the diagnosis of LM (odds ratio (OR) for LM 18.6; 95% confidence interval: 9.3-37.1).
  • The algorithm was equally effective in the diagnosis of amelanotic lesions and showed good interobserver reproducibility (87%).

  • [Title] Amelanotic esophageal malignant melanoma: case report and short review of the literature.
  • Malignant melanoma in the esophagus is a rare condition which has been described only occasionally in case reports or in larger series of patients with esophageal disease.
  • We describe here the very rare case of a patient who presented initially with a 2-month history of dysphagia and weight loss which led to the endoscopic diagnosis of an unclear lesion in the distal esophagus.
  • As there were no signs of skin melanoma and there was an absence of pigmentation, a diagnosis of primary amelanotic malignant melanoma was made.
  • At the end of May 2007, 4 months after the primary diagnosis, the patient died due to acute renal failure.

  • OBJECTIVE: To characterize a murine model of spontaneous amelanotic melanoma arising in the uvea of transgenic mice bearing a targeted deletion of the Ink4a/Arf tumor suppressor locus (exons 2 and 3) and expressing human H-ras controlled by the human tyrosinase promoter.
  • CONCLUSION: Spontaneous uveal malignant melanomas occurring in male Tyr-RAS+ Ink4a/Arf-/- transgenic mice arise within the choroid or ciliary body and share histopathological features characteristic of human uveal melanoma.
  • CLINICAL RELEVANCE: Uveal melanoma research has benefited from xenograft models, but engineered mouse models of spontaneous uveal amelanotic melanoma will undoubtedly further our understanding of the genetic underpinning for this disease.

  • Differential diagnoses included intraepidermal adenocarcinoma, in situ squamous or basal cell carcinoma, junctional amelanotic melanoma, and epitheliotropic tumours of histiocytic or lymphocytic origin.

  • BACKGROUND: The clinical diagnosis of amelanotic melanoma is still a challenge.
  • CASE REPORT: A 72-year-old patient was first referred to us for the excision of a melanoma of the chin.
  • In the first examination, we discovered another melanoma of the trunk.
  • Both malignancies were excised and histopathology confirmed the clinical diagnosis of melanomas with Clark level II and III, respectively.
  • Surprisingly, the pathology report showed typical features of a malignant melanoma.
  • The diagnosis was confirmed by 3 further punch biopsies (Clark level III, Breslow 0.6 mm).
  • CONCLUSIONS: Amelanotic melanoma may appear as an erythematous macula or plaque; clinical symptoms are erythema, edema, pruritus or a slow increase in size and hypopigmentation or discoloration.

  • Approximately, 1-4% of all new melanoma cases occur in patients younger than 20 years.
  • The clinical presentation of melanoma in the young is often challenging.
  • We report the experience of the Melanoma Unit of University Hospital Spedali Civili of Brescia, Italy.
  • Study subjects were drawn from 1470 patients with histologically confirmed melanoma.
  • From this group, melanoma developed in 12 patients younger than 19 years.
  • For each melanoma diagnosed, histologic characteristics, treatment, and outcomes were evaluated.
  • In many cases reported in literature lesions are amelanotic, nodular, and resemble pyogenic granuloma.
  • From our case studies it was found that the clinical characteristics detected in melanomas diagnosed in childhood and adolescence have been the same as those described in adults and that the ABCDE clinical criteria may be helpful basics of melanoma.

  • Melanoma has been reported to be susceptible to immune control, although the degree of impact of immunosuppression on the course of this disease is uncertain.
  • The Beth Israel Deaconess Medical Center's Cutaneous Oncology Program database was examined to identify patients receiving immunosuppressive therapy at the time of melanoma diagnosis.
  • Nineteen patients were identified with melanoma and concomitant immunosuppression in a database of 1839 patients.
  • Compared with the remaining database, patients on immunosuppression were more likely to be women (84 vs. 44%) with an amelanotic primary melanoma (21 vs. 5.3%).
  • Patients with immunosuppression were equally likely to relapse, but more likely to have died of melanoma than controls (42 vs. 23%) (P=0.01, log-rank test).
  • These findings suggest that immunosuppressive therapy is associated with a more aggressive disease course in patients with melanoma.
  • The additional observation that the stage-specific recurrence rates were similar, however, suggests that routine dermatologic screening of immunosuppressed patients may lead to earlier diagnosis and improved outcomes.

  • We report on a patient with rectal malignant melanoma.
  • With the diagnosis of neuroendocrine carcinoma of the rectum, abdominoperineal resection was performed.
  • However, the final pathological diagnosis of the surgically resected specimen was malignant amelanotic melanoma of the rectum.
  • As preoperative pathological diagnosis showed rare rectal tumor, we measured the chemosensitivity of the rectal tumor using the collagen gel droplet-embedded culture drug sensitivity test (CD-DST) to determine the most appropriate chemotherapy regimen for the patient.
  • However, there were no anticancer drugs tested by CD-DST for malignant melanoma.

  • Subungual melanoma is a disorder that accounts for 2% to 3% of all melanoma cases.
  • We report the case of a 31-year-old male patient presenting with a rapidly growing mass localized to the hallux, which eventually resulted in the diagnosis of an ulcerated amelanotic subungual melanoma.
  • Because of their lack of pigmentation, amelanotic melanoma is frequently misdiagnosed for other more common forms of cutaneous compromise, and a delay in proper treatment because of failure to make the diagnosis can greatly decrease the likelihood of a favorable outcome.

  • Subsequently, we confirmed the presence of red blood in stools, like enterorrhagia and underwent a colonoscopy, in which two elevated lesions were found in the cecum: a pedunculated (with active bleeding, oozing) and other sessile; both were removed. the pathology showed that corresponded to amelanotic melanoma in cecal region.

  • The aim of this study was to identify a phenolic prodrug compound that is minimally metabolized by rat liver microsomes, but yet could form quinone reactive intermediates in melanoma cells as a result of its bioactivation by tyrosinase.
  • In current work, we investigated 24 phenolic compounds for their metabolism by tyrosinase, rat liver microsomes and their toxicity towards murine B16-F0 and human SK-MEL-28 melanoma cells.
  • A linear correlation was found between toxicities of phenolic analogs towards SK-MEL-28 and B16-F0 melanoma cells, suggesting similar mechanisms of toxicity in both cell lines.
  • 4-HEB (IC(50) 48h, 75muM) showed selective toxicity towards five melanocytic melanoma cell lines SK-MEL-28, SK-MEL-5, MeWo, B16-F0 and B16-F10, which express functional tyrosinase, compared to four non-melanoma cells lines SW-620, Saos-2, PC3 and BJ cells and two amelanotic SK-MEL-24, C32 cells, which do not express functional tyrosinase.
  • Our findings suggest that presence of a phenolic group in 4-HEB is critical for its selective toxicity towards melanoma cells.

  • [Title] Does melanoma behave differently in younger children than in adults? A retrospective study of 33 cases of childhood melanoma from a single institution.
  • OBJECTIVE: To ascertain whether childhood melanoma presents any peculiar clinical features or differences in prognosis with respect to adults, we retrospectively analyzed the data from 33 patients who were up to 14 years of age and treated for cutaneous melanoma at the Istituto Nazionale Tumori, Milan, over a 25-year period.
  • METHODS: Primary lesions were amelanotic in half of the cases and raised in 73%.
  • Nine children had nodal involvement at diagnosis, 2 in-transit metastases, and 1 distant spread.
  • CONCLUSION: By comparison with adult cases, childhood melanoma can have a higher percentage of atypical clinical features (amelanotic and raised lesions), nodular histotype, and thick lesions.
  • Although we have no data to support any suggestion of biological differences between young children and adolescents or adults, our findings give the impression that melanoma behaves differently in the younger age group.

  • Collision (contiguous) tumors of the skin can result in misleading clinicopathological presentations, and the choice of appropriate diagnostic techniques may prevent incomplete diagnosis and management.
  • We report 2 cases of collision tumors involving amelanotic malignant melanoma of the back.
  • Subsequent excision showed that the lesion was largely composed of amelanotic melanoma underlying a relatively small and thin basal cell carcinoma, and this probably would have been demonstrated in a punch (rather than shave) biopsy.
  • The other patient is a 71-yr-old male with a 1 cm exophytic lesion on the back, which was determined microscopically to be melanoma, and a 0.6 cm papule on the back.
  • This lesion was composed of 2 distinct contiguous neoplastic infiltrates, the predominant component being an atypical fibroxanthoma and the smaller component an amelanotic melanoma (primary vs metastatic), with diagnostic confirmation requiring multiple immunohistochemical stains.

  • Loss of pigment in hamster amelanotic melanoma line is accompanied by a faster growth rate, higher tumorigenicity and shorter animal survival time.
  • Thus, the malignancy of melanoma increases during the alteration of melanotic (Ma) into amelanotic (Ab) line.
  • As changes in the ability to undergo a spontaneous or induced apoptosis, and the role of caspases in this process during melanoma progression are not well defined, they were investigated in this work.
  • Cytochrome c release into cytosol, and the activation of effector caspases, estimated by PARP degradation clearly showed that during the spontaneous death in the cells from both melanoma lines intrinsic way of apoptosis was activated.
  • Confocal and cytometric flow analyses indicate that camptothecin (CPT) induced apoptosis with caspase activation by the intrinsic way only in the amelanotic melanoma cells, even though cells of the Ma line also underwent CPT-induced apoptosis (the content of TUNEL-positive cells increased).
  • Thus, our results suggest that melanoma progression, associated with a decreased ability to undergo spontaneous apoptosis but an increased susceptibility to CPT-induced apoptosis, relates to different levels of caspase activation; they also show that intrinsic way of apoptotis depends on the phenotype of melanoma cells, being more pronounced in the melanotic melanoma cells.
  • On the other hand, melanotic melanoma cells resistance to camptothecin-induced apoptosis suggests that the melanogenic apparatus or melanin itself may have the protective effect on the ability of the melanoma cells to undergo apoptosis.

  • BACKGROUND: Acral lentiginous melanoma (ALM) is a rare but distinctive subtype of melanoma.
  • The diagnosis is often delayed and misdiagnosis is common, due to frequently unusual clinical presentation and a higher rate of amelanosis than in other melanoma subtypes.
  • In volar skin melanomas, the two most prevalent patterns were irregular diffuse pigmentation (60%) and the parallel-ridge pattern (53%).
  • The study included 37 (34%) amelanotic melanomas.
  • CONCLUSIONS: The presence of a parallel-ridge pattern and/or irregular diffuse pigmentation within the lesion is highly indicative of melanoma on volar skin.
  • Amelanotic ALM either in volar skin or in nail apparatus is characterized by remnants of pigmentation and a polymorphic vascular pattern.

  • This case report presents a very aggressive course of amelanotic nodular melanoma during pregnancy resulting in death five months after delivery.
  • A 34 year-old Caucasian woman at 19th week of the second pregnancy was diagnosed having amelanotic nodular melanoma (tumor thickness - 2.5 mm) with metastases to the regional right inguinal lymph node.
  • Amelanotic nodular melanoma represents malignant melanocytic tumor of the skin, which clinically mimics a variety of benign and malignant skin conditions and therefore commonly leads to delayed diagnosis.
  • Since pregnancy limits the prescription of immunotherapy and chemotherapy, the prognosis for melanoma during pregnancy detected later than in the second stage is poor and can be illustrated by our reported case.
  • Such patients seems to be at higher risk to develop metastasis of melanoma in the internal organs and occasionally even in the fetus; therefore, they should be timely informed about that.

  • Desmoplastic melanoma (DM) is a rare variant of spindle cell melanoma, which usually develops in sun-damaged skin of elderly patients.
  • Often the lesion is nonpigmented and frequently mistaken for a nonmelanocytic proliferation, which delays diagnosis and treatment and therefore worsens the prognosis.
  • Malignant melanoma was the initial clinical diagnosis in only 27% of the cases.
  • Fifty-one cases (45%) were classified as "pure DM" when the lesion was entirely desmoplastic, and 62 cases (55%) were considered as "combined DM" when a recognizable desmoplastic component was seen in an otherwise conventional malignant melanoma.
  • In 81% of the cases, an atypical intraepidermal melanocytic component (in situ malignant melanoma) was identified, whereas in the remaining 19% of the cases the intraepidermal component was lacking.
  • Seventy-one percent of the cases were histologically amelanotic, 23% showed a small amount of pigment, and only 6% were heavily pigmented.
  • Although a better prognosis has been postulated for DM when compared with conventional cutaneous malignant melanomas of the same thickness, in most cases, a DM is diagnosed only in established long-standing and thick melanomas.
  • Therefore, dermatologists and dermatopathologists should be more aware of this clinicopathologic variant of cutaneous malignant melanoma.

  • The most antiproliferative compounds against amelanotic melanoma cells were kaempferol-3-O-beta-D-glucopyranoside and irisolidone with a percentage of inhibition of 100 and 96.6, respectively, and against large lung carcinoma cells with a percentage of inhibition of 82.1 and 84.6, respectively.
  • Significant activity on the amelanotic melanoma cell line was also showed by irigenin-7-O-beta-D-glucopyranoside, with a percentage of inhibition of 89.3.
  • The compounds isovitexin and isoorientin-6-O''-beta-D-glucopyranoside showed a selective activity against amelanotic melanoma cells with a percentage of inhibition of 83.2 and 79.8, respectively.

  • In the present study, we examined the expression of the hair follicle and neural stem cell marker nestin, as well as S-100 and HMB-45, in melanoma.
  • Nestin immunoreactivity was observed in the HMB-45-negative melanoma cells in all five cases of amelanotic nodular melanomas.
  • Especially, nestin immunoreactivity was observed in the HMB-45-negative melanoma cells in the dermal parts of all 10 cases of HMB-45-negative amelanotic and melanotic nodular melanomas.
  • On the other hand, nestin expression was negative in 10 of 12 cases of superficial spreading melanoma.
  • These results suggest that nestin is an important marker of HMB-45-negative melanoma cells in the dermal parts of patients with nodular melanoma.

  • Dialkylamino-alkyl-benzamides possess an affinity for melanin, suggesting that labeling of such benzamides with (18)F could potentially produce melanin-targeted PET probes able to identify melanotic melanoma metastases in vivo with high sensitivity and specificity.
  • In vivo distribution and small-animal PET studies were conducted on mice bearing B16F10 melanoma, A375M amelanotic melanoma, and U87MG tumors, and comparative studies were performed with (18)F-FDG PET in the melanoma models.
  • In vivo, (18)F-FBZA displayed significant tumor uptake; at 2 h, 5.94 +/- 1.83 percentage injected dose (%ID) per gram was observed in B16F10 tumors and only 0.75 +/- 0.09 %ID/g and 0.56 +/- 0.13 %ID/g was observed in amelanotic A375M and U87MG tumors, respectively.
  • CONCLUSION: (18)F-FBZA specifically targets primary and metastatic melanotic melanoma lesions with high tumor uptake and may have translational potential.

  • Proteasomes have been shown to be involved in the regulation of melanin biosynthesis in melanoma cells.
  • Here we report on the correlation between proteasome subunits and Tyrosinase (Tyr) activity in different cell phenotypes, and thereby regulation of melanin biosynthesis in B16F10 mouse melanoma cells.
  • Our results indicated that the quantity of proteasome subunit p27 is higher and that of the enzyme Tyr and its activity are lower in amelanotic melanoma cells, while the reverse is true in melanotic melanoma cells.
  • Proteasome subunit p27, compared to another subunit p31, shows increased co-localization with Tyr and Tyrosinase related protein 1 (Trp1) in amelanotic cells to a greater extent than that in melanotic cells.
  • On exposure to cycloheximide, increased Tyr degradation was seen in amelanotic cells, as indicated by increased co-localization of p27 and Tyr.
  • Further, exposure of amelanotic melanoma cells with proteasome-specific inhibitor MG132 resulted in an increased Tyr activity, increased levels of Tyr and Trp1, leading to increased melanin synthesis.
  • These results therefore suggest that proteasomes, particularly p27 subunit, are directly involved in the regulation of melanin biosynthesis in mouse melanoma cells.

  • Antiproliferative activity was tested on amelanotic melanoma C32 cells and on renal cell adenocarcinoma cells, using the sulphorhodamine B assay.
  • RESULTS: Cupressus sempervirens ssp. pyramidalis leaf oil exerted the highest cytotoxic activity with an IC(50)value of 104.90 microg/mL against C32, followed by activity of P. orientalis and P. asperula on the renal adenocarcinoma cell line (IC(50) of 121.93 and 139.17 microg/mL, respectively). P. orientalis essential oil was also active against amelanotic melanoma with an IC(50) of 330.04 microg/mL.

  • This article provides a general overview of the dermoscopic evaluation of nonpigmented skin tumors and is divided into two parts.
  • Part I discusses the dermoscopic vascular patterns of benign and malignant melanocytic skin tumors.

  • According to data from the literature and to new data from the German central registry for malignant melanomas from the German Society of Dermatology, a summary is given about the occurrence of malignant melanoma in children and adolescents up to 18 years of age.
  • During the first 10 years of life the occurrence of a melanoma is extremely rare, but slowly increases after the first decade.
  • Several studies have shown that young people often have amelanotic tumors resembling pyogenic granuloma.
  • A reason might be a delay in the clinical diagnosis of malignant melanoma in children and young adults.
  • Clinical and histopathological examples of melanoma in young people are demonstrated.
  • Pitfalls in differential diagnosis and simulators of malignant melanoma are described and illustrated by examples of a medium sized congenital nevus with extensive intraepithelial spread of single melanocytes resembling a superficial spreading melanoma and by a compound nevus in association with a lichen sclerosus et atrophicus.

  • Pigments serve many visually obvious animal functions (e.g. hair, skin, eyes, feathers, scales).
  • Clinicians and researchers are well acquainted with melanin in skin and ocular pathologies and now increasingly are with internal, melanized, pathology-associated sites not obviously subject to light radiation (e.g. brain, cochlea).
  • At both types of sites some findings puzzle: positive and negative neuromelanin effects in Parkinsons; unexpected melanocyte action in the cochlea, in deafness; melanin reduces DNA damage, but can promote melanoma; in melanotic cells, mitochondrial number was 83% less, respiration down 30%, but development similar to normal amelanotic cells.

  • OBSERVATIONS: We describe 3 cases of in situ amelanotic melanoma of the nail unit.

  • Two patients had a history of malignant, non-ocular, neuroendocrine neoplasms (Merkel cell carcinoma and lung carcinoid in one patient each).
  • RESULTS: Ophthalmic examination revealed an amelanotic ciliochoroidal mass in 2 cases and a lightly melanotic and a pale orange choroidal mass in 1 case each.
  • Our differential diagnosis in all cases was metastatic carcinoma versus primary uveal melanoma.
  • FNAB of the intraocular mass was performed in all patients to establish a pathologic diagnosis and guide subsequent management.
  • Our final diagnosis was metastatic lung carcinoid in 2 patients and metastatic Merkel cell carcinoma and small cell lung carcinoma in 1 patient each.
  • CONCLUSIONS: Neuroendocrine tumours are a heterogeneous group of neoplasms whose diagnosis ultimately depends on the identification of specific cell markers (e.g., neuron-specific enolase, chromogranin, synaptophysin), hormones and neurotransmitters (e.g., gastrin, serotonin, adrenocorticotrophic hormone [ACTH]).
  • FNAB with immunohistochemical stains for neuroendocrine markers can establish a pathologic diagnosis in cases of uveal metastasis from a neuroendocrine tumour.

  • BACKGROUND: The amelanotic melanoma of the rectal mucosa is very rare with an unfavourable prognosis.
  • Therefore transrectal ultrasound is of major importance in the preoperative staging and postoperative follow-up especially in diagnosis of local recurrence by using the ultrasound-guided, transrectal aspiration.
  • METHODS: In literature 5 cases of amelanotic malignant melanoma were reported.
  • The overall survival time is 10 months after diagnosis.
  • RESULTS: We report about a 55-year-old female patient with an amelanotic melanoma of rectal mucosa.
  • The patient died 36 months after diagnosis.
  • CONCLUSION: The prognosis of primary malignant anorectal melanoma is poor, irrespective of surgical treatment.
  • Wide local resection is the first choice for primary anorectal melanoma.
  • Chemotherapy, radiotherapy and immunotherapy should be considered in the treatment of anorectal melanoma to influence the overall survival.

  • The efficiency of a Zn(II)-phthalocyanine disulphide (C11Pc), a compound with both phthalocyanine units bearing seven hexyl chains and a sulphur terminated C11 chain, as a photodynamic therapy (PDT) agent was investigated in C57 mice bearing a sub-cutaneously transplanted amelanotic melanoma.
  • Biodistribution studies at selected post-injection times showed that the nanoparticle-associated C11Pc was recovered in significantly larger amounts from all the examined tissues and the serum and yielded a greater selectivity of tumour targeting: thus, the ratio between the amount of phthalocyanine recovered from the amelanotic melanoma and the skin (peritumoural tissue) increased from 2.3 to 5.5 from the free to the gold nanoparticle-bound C11Pc at 24 h after injection.
  • PDT studies with the C11Pc-loaded amelanotic melanoma showed a markedly more significant response of the tumour in the mice that had received the nanoparticle-bound photosensitiser; the PDT effect was especially extensive if the irradiation was performed at 3h after C11Pc injection when large phthalocyanine amounts were still present in the serum.

  • DIAGNOSIS: The autopsy revealed that he was affected by two malignant neoplasms simultaneously: an amelanotic malignant melanoma metastasizing into a localized renal cell carcinoma.

  • CASE REPORT: We describe the clinical presentation of a choroidal amelanotic melanoma in a 46-year-old woman with oculocutaneous albinism.
  • Microscopic findings and histopathological features, demonstrating a spindle B-cell melanoma of the choroid, are also reviewed.
  • COMMENTS: Magnetic resonance imaging may be helpful in diagnosing amelanotic melanoma in patients with oculocutaneous albinism.

  • [Title] Anti-tumor action of tumor necrosis factor against Bomirski Ab melanoma in hamsters.
  • MATERIALS AND METHODS: The effects of intratumoral injection of rat TNF into hamsters bearing Bomirski Ab amelanotic melanoma, a fast growing tumor of high metastatic potential, were tested.
  • In vitro assays indicated a direct cytotoxic effect of TNF against Ab melanoma cells, most probably as an outcome of apoptosis.
  • Intratumoral application of TNF also caused some modulation of cytokine response in melanoma-bearing hamsters as evidenced by increased levels of IL-6 in blood serum.
  • CONCLUSIONS: This study established Bomirski Ab melanoma as a useful model for complex analysis of the anti-tumor activity of TNF.

  • [Title] Melanosomal damage in normal human melanocytes induced by UVB and metal uptake--a basis for the pro-oxidant state of melanoma.
  • Melanins are ubiquitous catecholic pigments, formed in organelles called melanosomes within melanocytes, the function of which is to protect skin against harmful effects of UV radiation.
  • Melanosomes within melanoma cells are characteristically abnormal, with fragmented melanin and disrupted membranes.
  • We hypothesize that the disruption of melanosomal melanin might be an early event in the etiology and progression of melanoma, leading to increased oxidative stress and mutation.
  • Single-dose UVB/Cu treatment does not greatly affect cell viability or cell cycle progression in heavily pigmented cells, but did so in an amelanotic early stage melanoma cell line.

  • BACKGROUND: Amelanotic melanomas (AM) are a difficult diagnostic challenge for clinicians.
  • Sex, age, the clinical features, the site of presentation, the suspected diagnosis, the clinical course, the histological type, the Clark level and the Breslow thickness were recorded.
  • AM were divided in three main clinical types: an erythematous macule or patch on sun-exposed skin, a dermal plaque or nodule without a particular epidermal change, an exophytic nodule.
  • Histological subtypes considered were superficial spreading melanoma, nodular melanoma, and lentigo maligna melanoma.
  • Diagnostic delay considered from when the patients first noticed the lesion on the site where the melanoma was diagnosed and when the physician or the patient first proposed the removal was recorded.
  • The diagnostic delay did not differ between amelanotic and pigmented melanomas, nor between nodular AM and nodular pigmented melanomas.

  • [Title] Anorectal amelanotic melanoma.
  • OBJECTIVE: Anorectal melanoma is a rare, highly malignant tumour with a poor 5 year survival of 10%.
  • Most anorectal melanomas have gross and/or histologic pigmentation, however about 30% of anorectal melanomas are amelanotic.
  • METHOD: We report three cases of amelanotic anorectal melanomas and integrate our data with six case reports of amelanotic malignant melanoma from the literature.
  • Further we compare clinicopathological data and clinical outcome with large series of anorectal melanomas (both, amelanotic and pigmentated).
  • Rectal bleeding was the leading symptom in all cases with a mean duration of 4 months before diagnosis.
  • CONCLUSION: Early diagnosis is key for efficient treatment and improved survival rate for patients with this unusual variant of melanoma.
  • There is no difference in terms of age, time of diagnosis, stage and survival between pigmented and amelanotic anorectal melanoma.

  • [Title] CDKN2A and MC1R analysis in amelanotic and pigmented melanoma.
  • Amelanotic melanoma (AM) is a rare subtype of melanoma with little or no clinically visible pigment; it is more difficult to diagnose than pigmented melanoma (PM), and has a worse prognosis.
  • In the attempt to find a genetic explanation for the distinction between AM and PM, we conducted a case-case study, matching AM and PM patients, and testing them for germline mutations in high- (p16INK4A, p14ARF, CDK4) and low-penetrance (MC1R) melanoma susceptibility genes.
  • This rare mutation, which has been described only once previously, may be involved in predisposition to the amelanotic phenotype in combination with germline MC1R variants and coordinate somatic expression of pigmentation genes and their regulators.

  • [Title] A Case of Amelanotic Subungual Melanoma.
  • Amelanotic subungual melanoma is a rare dermatosis, and it is frequently misdiagnosed probably because of its nonspecific clinical features.
  • We herein report on a case of amelanotic subungual melanoma extended to the adjacent skin in a 36-year-old Korean woman.
  • This case is interesting in that clinically, it needed differentiation from Bowen's disease, lichen planus, sarcoidosis, etc. and very early invading features of the melanoma were observed on the histopathologic section.
