  • [Title] Fas and FasL expression on cells of two transplantable melanoma lines according to their different biological properties.
  • In tumours (e.g. melanoma), FasL expression possibly counteracts the Fas-positive effector T cells that infiltrate into tumours, and consequently the Fas/FasL interaction can contribute to the escape of tumour cells from the systemic immune response.
  • In this study we examined differences in Fas and FasL expression on cells from the hamster melanotic melanoma line (Ma) and a more aggressive amelanotic melanoma line (Ab).
  • We also tried to find out whether the Fas/FasL expression induces an ability to undergo spontaneous apoptosis in these two transplantable melanoma lines.
  • Isolated transplantable melanoma cells were incubated for 4 and 24 hours and after that time the expression of Fas and FasL was estimated by flow cytometry.
  • The results show that there was no Fas expression, although FasL was detected on both melanoma cell lines.
  • Therefore the data reported by other authors indicate that a lack or a low level of Fas expression and an ectopic expression of FasL on melanoma cells can be an escape mechanism of the tumour, to avoid host immune responses.
  • The content of FasL-positive melanotic melanoma cells was higher than in amelanotic melanoma cells and increased with the prolongation of the incubation time.
  • FasL expression on amelanotic melanoma cells was detected after 24 hours at a level similar to that on melanotic melanoma cells after 4 hours incubation time.
  • FasL expression on melanoma cells can induce apoptosis in cytotoxic T lymphocytes and NK cells which are responsible for tumour cells elimination.
  • The results obtained suggest that the Fas/FasL system does not play any significant role in spontaneous apoptosis of two melanoma cell lines.

  • Melanoma metastasizing to the lungs is common, but primary pulmonary or pleural melanoma is extremely rare.
  • We present an autopsy case of malignant melanoma of the pleura without primary skin lesion in a 49-year-old man.
  • At autopsy, a yellow-white tumor located primarily in the right visceral pleura (diagnosed as an amelanotic melanoma) was found to have invaded into the right lung, right parietal pleura, and right diaphragm, and to have metastasized into the left lung and visceral pleura, thyroid, and left adrenal gland.
  • Immuno-histochemical examination for S100 and HMB-45 would thus appear to be useful for the diagnosis of an amelanotic melanoma.

  • BACKGROUND: Primary malignant melanoma of the vagina is extremely rare, accounting for 0.3-0.8% of all malignant melanomas.
  • True amelanotic vaginal melanoma showing no melanin on histological examination is exceedingly rare, accounting for only 2% of all vaginal melanomas.
  • CASE REPORT: We describe a 31-year-old female patient who presented with locally advanced amelanotic melanoma of the vagina, with no evidence of metastatic spread on the computerized tomography (CT) scan, but who was subsequently diagnosed as suffering from metastatic disease by positron emission tomography (PET)-CT performed a few weeks following posterior pelvic exenteration.
  • CONCLUSION: Specific immunohistochemical staining with melanoma markers should be performed to confirm or exclude a diagnosis of amelanotic melanoma in all patients presenting with a vaginal mass composed of undifferentiated epithelioid malignant cells.
  • Fluorodeoxyglucose (FDG)-PET-CT should be performed as part of the preoperative evaluation, to identify the presence or absence of metastatic disease in all patients with vaginal melanoma.

  • BACKGROUND: The clinical diagnosis of amelanotic melanoma is still a challenge.
  • CASE REPORT: A 72-year-old patient was first referred to us for the excision of a melanoma of the chin.
  • In the first examination, we discovered another melanoma of the trunk.
  • Both malignancies were excised and histopathology confirmed the clinical diagnosis of melanomas with Clark level II and III, respectively.
  • Surprisingly, the pathology report showed typical features of a malignant melanoma.
  • The diagnosis was confirmed by 3 further punch biopsies (Clark level III, Breslow 0.6 mm).
  • CONCLUSIONS: Amelanotic melanoma may appear as an erythematous macula or plaque; clinical symptoms are erythema, edema, pruritus or a slow increase in size and hypopigmentation or discoloration.

  • Past history was unremarkable for skin lesions, biopsy or any surgery.
  • The lump was hard and freely mobile with normal overlying skin, nipple, and areola.
  • Careful examination of the skin did not reveal any lesion.
  • Immunoreactivity for S-100 and HMB-45, confirmed the diagnosis of amelanotic melanoma.
  • It is important to be aware of this atypical presentation of amelanotic melanoma in the breast and to utilize immunohistochemistry to differentiate from other commoner high-grade malignant tumors.

  • Two patients had a history of malignant, non-ocular, neuroendocrine neoplasms (Merkel cell carcinoma and lung carcinoid in one patient each).
  • RESULTS: Ophthalmic examination revealed an amelanotic ciliochoroidal mass in 2 cases and a lightly melanotic and a pale orange choroidal mass in 1 case each.
  • Our differential diagnosis in all cases was metastatic carcinoma versus primary uveal melanoma.
  • FNAB of the intraocular mass was performed in all patients to establish a pathologic diagnosis and guide subsequent management.
  • Our final diagnosis was metastatic lung carcinoid in 2 patients and metastatic Merkel cell carcinoma and small cell lung carcinoma in 1 patient each.
  • CONCLUSIONS: Neuroendocrine tumours are a heterogeneous group of neoplasms whose diagnosis ultimately depends on the identification of specific cell markers (e.g., neuron-specific enolase, chromogranin, synaptophysin), hormones and neurotransmitters (e.g., gastrin, serotonin, adrenocorticotrophic hormone [ACTH]).
  • FNAB with immunohistochemical stains for neuroendocrine markers can establish a pathologic diagnosis in cases of uveal metastasis from a neuroendocrine tumour.

  • OBJECTIVE: To characterize a murine model of spontaneous amelanotic melanoma arising in the uvea of transgenic mice bearing a targeted deletion of the Ink4a/Arf tumor suppressor locus (exons 2 and 3) and expressing human H-ras controlled by the human tyrosinase promoter.
  • CONCLUSION: Spontaneous uveal malignant melanomas occurring in male Tyr-RAS+ Ink4a/Arf-/- transgenic mice arise within the choroid or ciliary body and share histopathological features characteristic of human uveal melanoma.
  • CLINICAL RELEVANCE: Uveal melanoma research has benefited from xenograft models, but engineered mouse models of spontaneous uveal amelanotic melanoma will undoubtedly further our understanding of the genetic underpinning for this disease.

  • In tyrosinase-positive amelanotic melanomas this rate limiting enzyme is inactive because of acidic endo-melanosomal pH.
  • METHODS: The expression of the HPV16-E5 oncoproteins was induced in two Tyrosinase-positive amelanotic melanomas (the cell lines FRM and M14) by a retroviral expression construct.
  • In amelanotic melanomas these effects can modulate the cell phenotype and can induce a higher sensitivity to tyrosine related anti-blastic drugs.

  • Irradiation of B78H1 murine amelanotic melanoma cells with 850 nm light emitted from a Ti:sapphire laser, operated in a pulsed mode at high fluence rates and in the presence of Ni(II)-octabutoxy-naphthalocyanine (NiNc), promoted a photothermally sensitised process leading to fast and irreversible cell death.
  • Very similar results were obtained upon irradiation of NiNc-loaded C32 human amelanotic melanoma cells and transformed murine HT-1080 and HaCaT fibroblasts.
  • From these results, photothermal sensitisation appears to be a general phenomenon and preliminary studies with mice bearing subcutaneously transplanted amelanotic melanomas, irradiated with 850 nm light 24 h after intravenous injection of NiNc, suggest that this approach has potential for the therapy of some types of skin tumours.

  • Loss of pigment in hamster amelanotic melanoma line is accompanied by a faster growth rate, higher tumorigenicity and shorter animal survival time.
  • Thus, the malignancy of melanoma increases during the alteration of melanotic (Ma) into amelanotic (Ab) line.
  • As changes in the ability to undergo a spontaneous or induced apoptosis, and the role of caspases in this process during melanoma progression are not well defined, they were investigated in this work.
  • Cytochrome c release into cytosol, and the activation of effector caspases, estimated by PARP degradation clearly showed that during the spontaneous death in the cells from both melanoma lines intrinsic way of apoptosis was activated.
  • Confocal and cytometric flow analyses indicate that camptothecin (CPT) induced apoptosis with caspase activation by the intrinsic way only in the amelanotic melanoma cells, even though cells of the Ma line also underwent CPT-induced apoptosis (the content of TUNEL-positive cells increased).
  • Thus, our results suggest that melanoma progression, associated with a decreased ability to undergo spontaneous apoptosis but an increased susceptibility to CPT-induced apoptosis, relates to different levels of caspase activation; they also show that intrinsic way of apoptotis depends on the phenotype of melanoma cells, being more pronounced in the melanotic melanoma cells.
  • On the other hand, melanotic melanoma cells resistance to camptothecin-induced apoptosis suggests that the melanogenic apparatus or melanin itself may have the protective effect on the ability of the melanoma cells to undergo apoptosis.

  • We report on a patient with rectal malignant melanoma.
  • With the diagnosis of neuroendocrine carcinoma of the rectum, abdominoperineal resection was performed.
  • However, the final pathological diagnosis of the surgically resected specimen was malignant amelanotic melanoma of the rectum.
  • As preoperative pathological diagnosis showed rare rectal tumor, we measured the chemosensitivity of the rectal tumor using the collagen gel droplet-embedded culture drug sensitivity test (CD-DST) to determine the most appropriate chemotherapy regimen for the patient.
  • However, there were no anticancer drugs tested by CD-DST for malignant melanoma.

  • BACKGROUND: Nail apparatus melanoma is known to be associated with a poor prognosis, mainly because of a delay in diagnosis that is made at an invasive stage.
  • This delay is particularly true in cases involving amelanotic melanoma.
  • To our knowledge, only 1 case of in situ amelanotic melanoma of the nail unit has previously been described.
  • We report 3 cases of in situ amelanotic melanoma with clinical lichenoid features.
  • OBSERVATIONS: We describe 3 cases of in situ amelanotic melanoma of the nail unit.
  • Histologic examination revealed in situ amelanotic melanoma extending from the proximal matrix up to the distal part of the nail bed.
  • CONCLUSIONS: Monodactylic lichenoid nail changes should be added to the more conventional signs of incipient nail melanoma.

  • [Title] Intracranial trigeminal nerve metastasis of a desmoplastic neurotropic melanoma: case report.
  • BACKGROUND: Desmoplastic neurotropic mela-noma is a rare and highly malignant variant of melanoma.
  • Solitary nervus trigeminus and Gasserian ganglion metastasis of a neurotropic melanoma has not been previously described in the literature.
  • 4 years previously he underwent tumor removal with an initial diagnosis of amelanotic malignant cutaneous melanoma; 1 year later, because of tumor recurrence, the patient underwent neck dissection, chemotherapy and radiation.
  • Histopathological studies proved the tumor to be a desmoplastic neurotropic melanoma (DNM) that was related to the previously treated malignant melanoma.
  • CONCLUSION: A metastatic tumor arising solely in a trigeminal nerve from a cutaneous malignant melanoma is quite rare; to our knowledge this may be the first report of such a case in the literature.

  • BACKGROUND: Amelanotic melanoma can mimic a wide variety of epithelial and nonepithelial malignant tumors.
  • Varied cytomorphology of melanoma has been described on exfoliative and fine needle aspiration cytology (FNAC).
  • We report a case of recurrent amelanotic melanoma to highlight its varied cytomorphologic features, which may cause diagnostic problems on cytologic and on histologic examinations.
  • FNAC revealed malignant cells with highly varied morphology with plasmacytoid and pleomorphic malignant cells with occasional fibrocollagenous tissue strands showing adherent neoplastic cells.
  • A cytologic diagnosis of pleomorphic malignant tumor was suggested, and the original histologic slides were reviewed; they showed a striking alveolar pattern that vaguely resembled an alveolar rhabdomyosarcoma.
  • A final diagnosis of amelanotic melanoma was made.
  • CONCLUSION: Awareness of the highly varied cytomorphology of amelanotic melanoma minimizes the diagnostic difficulty on fine needle aspiration smears.

  • STUDY DESIGN/MATERIALS AND METHODS: Two octabutoxy-naphthalocyanines centrally coordinated with Pd(II) or Pt(II) ions were prepared by chemical synthesis and tested for their affinity and photothermal sensitisation activity toward a selected tumour cell line, namely B78H1 amelanotic melanoma.
  • The results thus obtained provided a basis for subsequent in vivo studies, aimed at defining the phototherapeutic efficiency of the two metallo-naphthalocyanines: the photosensitisers were i.v. injected into C57BL/6 mice bearing a subcutaneously transplanted amelanotic melanoma and at 24 hours post-injection the tumour area was irradiated by the Ti:sapphire laser using the same protocol as above detailed.
  • RESULTS: Both naphthalocyanines exhibited a high affinity for the amelanotic melanoma cells.
  • Lastly, both metallo-naphthalocyanines, and in particular the Pd(II) derivative, promoted an important response by the amelanotic melanoma, when the neoplastic tissue was irradiated by the pulsed Ti:sapphire laser.

  • We have previously shown that the DRO subline of the amelanotic melanoma A375 responds to rexinoid and thiazolidinedione (TZD) treatment in vitro and in vivo.

  • According to data from the literature and to new data from the German central registry for malignant melanomas from the German Society of Dermatology, a summary is given about the occurrence of malignant melanoma in children and adolescents up to 18 years of age.
  • During the first 10 years of life the occurrence of a melanoma is extremely rare, but slowly increases after the first decade.
  • Several studies have shown that young people often have amelanotic tumors resembling pyogenic granuloma.
  • A reason might be a delay in the clinical diagnosis of malignant melanoma in children and young adults.
  • Clinical and histopathological examples of melanoma in young people are demonstrated.
  • Pitfalls in differential diagnosis and simulators of malignant melanoma are described and illustrated by examples of a medium sized congenital nevus with extensive intraepithelial spread of single melanocytes resembling a superficial spreading melanoma and by a compound nevus in association with a lichen sclerosus et atrophicus.

  • BACKGROUND: Different dermatoscopic algorithms have been developed to evaluate pigmented lesions of the skin, with pattern analysis being the most commonly used.
  • We sought to develop and evaluate a diagnostic scoring system, the reticular point of view, to distinguish common melanocytic nevus from dysplastic nevus and from melanoma.
  • Regarding the diagnosis of melanoma, only the presence of large holes, areas of abrupt cut off, and linear extensions revealed statistical significance.
  • It is not applicable in nodular, thick, and amelanotic melanomas that are usually lacking in pigment network.
  • CONCLUSION: Although pattern analysis represents the most effective analytical method in dermatoscopy, our scoring system may be useful to distinguish between benign lesions, which need only clinical follow-up, and malignant lesions, which need surgical excision.

  • OBJECTIVE: To examine the significance of eccentric hyperpigmentation (EH), central hyperpigmentation (CH), multifocal hyper/hypopigmentation (MH/HP), and the multicomponent pattern (MCP) in melanocytic lesions lacking specific dermoscopic features of melanoma.
  • DESIGN: A total of 3367 benign and malignant melanocytic lesions (n = 341 melanomas, excluding lentigo maligna and lentigo maligna melanoma) were examined to identify those lesions lacking specific dermoscopic features of melanoma but having any of the global patterns of EH, CH, MH/HP, and MCP.
  • SETTING: Dermoscopic images were collected from lesions excised or undergoing sequential digital monitoring from the Sydney Melanoma Diagnostic Centre, a tertiary referral institution located in Sydney, Australia.
  • MAIN OUTCOME MEASURE: The odds ratio (OR) for melanoma of EH, CH, MH/HP, and MCP.
  • RESULTS: While EH (OR, 3.3; 95% confidence interval [CI], 2.5-4.6) and MCP (OR, 15.4; 95% CI, 11.9-19.9) were significant predictors of melanoma when total melanomas vs nevi were analyzed, there was no significant difference between the frequency of any of the global patterns in melanomas vs benign nevi lacking specific dermoscopic features of melanoma.
  • CONCLUSION: Based on our study results and previous prevalence data on these global patterns in benign nevi, we do not believe that lesions with EH or MCP require closer observation than other benign nevi lacking specific dermoscopic features of melanoma.

  • The most antiproliferative compounds against amelanotic melanoma cells were kaempferol-3-O-beta-D-glucopyranoside and irisolidone with a percentage of inhibition of 100 and 96.6, respectively, and against large lung carcinoma cells with a percentage of inhibition of 82.1 and 84.6, respectively.
  • Significant activity on the amelanotic melanoma cell line was also showed by irigenin-7-O-beta-D-glucopyranoside, with a percentage of inhibition of 89.3.
  • The compounds isovitexin and isoorientin-6-O''-beta-D-glucopyranoside showed a selective activity against amelanotic melanoma cells with a percentage of inhibition of 83.2 and 79.8, respectively.

  • Dialkylamino-alkyl-benzamides possess an affinity for melanin, suggesting that labeling of such benzamides with (18)F could potentially produce melanin-targeted PET probes able to identify melanotic melanoma metastases in vivo with high sensitivity and specificity.
  • In vivo distribution and small-animal PET studies were conducted on mice bearing B16F10 melanoma, A375M amelanotic melanoma, and U87MG tumors, and comparative studies were performed with (18)F-FDG PET in the melanoma models.
  • In vivo, (18)F-FBZA displayed significant tumor uptake; at 2 h, 5.94 +/- 1.83 percentage injected dose (%ID) per gram was observed in B16F10 tumors and only 0.75 +/- 0.09 %ID/g and 0.56 +/- 0.13 %ID/g was observed in amelanotic A375M and U87MG tumors, respectively.
  • CONCLUSION: (18)F-FBZA specifically targets primary and metastatic melanotic melanoma lesions with high tumor uptake and may have translational potential.

  • Proteasomes have been shown to be involved in the regulation of melanin biosynthesis in melanoma cells.
  • Here we report on the correlation between proteasome subunits and Tyrosinase (Tyr) activity in different cell phenotypes, and thereby regulation of melanin biosynthesis in B16F10 mouse melanoma cells.
  • Our results indicated that the quantity of proteasome subunit p27 is higher and that of the enzyme Tyr and its activity are lower in amelanotic melanoma cells, while the reverse is true in melanotic melanoma cells.
  • Proteasome subunit p27, compared to another subunit p31, shows increased co-localization with Tyr and Tyrosinase related protein 1 (Trp1) in amelanotic cells to a greater extent than that in melanotic cells.
  • On exposure to cycloheximide, increased Tyr degradation was seen in amelanotic cells, as indicated by increased co-localization of p27 and Tyr.
  • Further, exposure of amelanotic melanoma cells with proteasome-specific inhibitor MG132 resulted in an increased Tyr activity, increased levels of Tyr and Trp1, leading to increased melanin synthesis.
  • These results therefore suggest that proteasomes, particularly p27 subunit, are directly involved in the regulation of melanin biosynthesis in mouse melanoma cells.

  • Herein, we report a rare case of amelanotic spindle-cell melanoma on the interdigit of the left fifth toe of an 83-year-old woman.
  • Two biopsies could not lead us to the correct diagnosis until the totally excised specimen was evaluated with immunohistochemical analysis including S-100 and other melanocyte markers.
  • This case suggests that refractory interdigital dermatophytoses should be treated by considering the possibility of concomitant malignant neoplasms, and immunohistochemical analysis is indispensable for differential diagnosis of malignant neoplasms suggesting nonspecific granulation.

  • The study concerns DNA ploidy and proliferative activity of the cells of two hamster transplantable melanoma lines - differing in many biological features - in the light of possible changes in their secretory activity.
  • Our results indicate that melanotic melanoma cells (Ma) have a near-tetraploid DNAcontent and about 18% of proliferating cells, while amelanotic melanoma cells (Ab) - have a near-triploid DNA content and almost twice as many proliferating cells.
  • The Ab cells, in comparison with Ma cells, secreted in vitro less total protein and most of the cytokines examined except OSM, but a spontaneous alteration of transplantable melanoma was accompanied by an increase of the quantity and dynamics of NO secretion.
  • So, the cells of two melanoma lines have their own characteristic pattern of secretory function.
  • But, the aneuploidy which accompanied the changes in cell differentiation of the studied melanoma lines, although seemed to reflect their changes in the proliferative activity, nevertheless did not reflect, in a direct way, differences in the secretion of the substances studied.

  • [Title] The in vivo efficacy of phthalocyanine-nanoparticle conjugates for the photodynamic therapy of amelanotic melanoma.
  • The efficiency of a Zn(II)-phthalocyanine disulphide (C11Pc), a compound with both phthalocyanine units bearing seven hexyl chains and a sulphur terminated C11 chain, as a photodynamic therapy (PDT) agent was investigated in C57 mice bearing a sub-cutaneously transplanted amelanotic melanoma.
  • Biodistribution studies at selected post-injection times showed that the nanoparticle-associated C11Pc was recovered in significantly larger amounts from all the examined tissues and the serum and yielded a greater selectivity of tumour targeting: thus, the ratio between the amount of phthalocyanine recovered from the amelanotic melanoma and the skin (peritumoural tissue) increased from 2.3 to 5.5 from the free to the gold nanoparticle-bound C11Pc at 24 h after injection.
  • PDT studies with the C11Pc-loaded amelanotic melanoma showed a markedly more significant response of the tumour in the mice that had received the nanoparticle-bound photosensitiser; the PDT effect was especially extensive if the irradiation was performed at 3h after C11Pc injection when large phthalocyanine amounts were still present in the serum.

  • We present a case of "true" amelanotic melanoma on the forehead of an 89-year-old man, which clinically mimicked squamous cell carcinoma.

  • Subungual melanoma is a disorder that accounts for 2% to 3% of all melanoma cases.
  • We report the case of a 31-year-old male patient presenting with a rapidly growing mass localized to the hallux, which eventually resulted in the diagnosis of an ulcerated amelanotic subungual melanoma.
  • Because of their lack of pigmentation, amelanotic melanoma is frequently misdiagnosed for other more common forms of cutaneous compromise, and a delay in proper treatment because of failure to make the diagnosis can greatly decrease the likelihood of a favorable outcome.

  • Desmoplastic melanoma (DM) is a rare variant of spindle cell melanoma, which usually develops in sun-damaged skin of elderly patients.
  • Often the lesion is nonpigmented and frequently mistaken for a nonmelanocytic proliferation, which delays diagnosis and treatment and therefore worsens the prognosis.
  • Malignant melanoma was the initial clinical diagnosis in only 27% of the cases.
  • Fifty-one cases (45%) were classified as "pure DM" when the lesion was entirely desmoplastic, and 62 cases (55%) were considered as "combined DM" when a recognizable desmoplastic component was seen in an otherwise conventional malignant melanoma.
  • In 81% of the cases, an atypical intraepidermal melanocytic component (in situ malignant melanoma) was identified, whereas in the remaining 19% of the cases the intraepidermal component was lacking.
  • Seventy-one percent of the cases were histologically amelanotic, 23% showed a small amount of pigment, and only 6% were heavily pigmented.
  • Although a better prognosis has been postulated for DM when compared with conventional cutaneous malignant melanomas of the same thickness, in most cases, a DM is diagnosed only in established long-standing and thick melanomas.
  • Therefore, dermatologists and dermatopathologists should be more aware of this clinicopathologic variant of cutaneous malignant melanoma.

  • Hereditary sensory neuropathy type I (HSN I) is a slowly progressive neurological disorder characterised by prominent predominantly distal sensory loss, autonomic disturbances, autosomal dominant inheritance, and juvenile or adulthood disease onset.
  • Variable distal muscle weakness and wasting, and chronic skin ulcers are characteristic.
  • Diagnosis is based on the clinical observation and is supported by a family history.
  • Definitive diagnosis is based on the detection of mutations by direct sequencing of the SPTLC1 and RAB7 genes.
  • Correct clinical assessment and genetic confirmation of the diagnosis are important for appropriate genetic counselling and prognosis.
  • Differential diagnosis includes the other hereditary sensory and autonomic neuropathies (HSAN), especially HSAN II, as well as diabetic foot syndrome, alcoholic neuropathy, neuropathies caused by other neurotoxins/drugs, immune mediated neuropathy, amyloidosis, spinal cord diseases, tabes dorsalis, lepra neuropathy, or decaying skin tumours like amelanotic melanoma.

  • BACKGROUND: The amelanotic melanoma of the rectal mucosa is very rare with an unfavourable prognosis.
  • Therefore transrectal ultrasound is of major importance in the preoperative staging and postoperative follow-up especially in diagnosis of local recurrence by using the ultrasound-guided, transrectal aspiration.
  • METHODS: In literature 5 cases of amelanotic malignant melanoma were reported.
  • The overall survival time is 10 months after diagnosis.
  • RESULTS: We report about a 55-year-old female patient with an amelanotic melanoma of rectal mucosa.
  • The patient died 36 months after diagnosis.
  • CONCLUSION: The prognosis of primary malignant anorectal melanoma is poor, irrespective of surgical treatment.
  • Wide local resection is the first choice for primary anorectal melanoma.
  • Chemotherapy, radiotherapy and immunotherapy should be considered in the treatment of anorectal melanoma to influence the overall survival.

  • Melanoma accounts for only about 4% of all skin cancer cases but most of skin cancer-related deaths.
  • Standard systemic therapies such as interferon (IFN) have not been adequately effective in the management of melanoma.
  • Chemoprevention by naturally occurring agents present in food and beverages has shown benefits in certain cancers including nonmelanoma skin cancers.
  • Here, employing 2 human melanoma cell lines (A-375 amelanotic malignant melanoma and Hs-294T metastatic melanoma) and normal human epidermal melanocytes (NHEM), we studied the antiproliferative effects of epigallocatechin-3-gallate (EGCG), the major polyphenolic antioxidant present in green tea.
  • EGCG treatment was found to result in a dose-dependent decrease in the viability and growth of both melanoma cell lines.
  • EGCG treatment of the melanoma cell lines resulted in decreased cell proliferation (as assessed by Ki-67 and PCNA protein levels) and induction of apoptosis (as assessed cleavage of PARP, TUNEL assay and JC-1 assay).
  • EGCG also significantly inhibited the colony formation ability of the melanoma cells studied.
  • EGCG treatment of melanoma cells resulted in a downmodulation of anti-apoptotic protein Bcl2, upregulation of proapoptotic Bax and activation of caspases -3, -7 and -9.
  • Our data suggest that EGCG causes significant induction of cell cycle arrest and apoptosis of melanoma cells that is mediated via modulations in the cki-cyclin-cdk network and Bcl2 family proteins.
  • Thus, EGCG, alone or in conjunction with current therapies, could be useful for the management of melanoma.

  • MATERIALS AND METHODS: The effects of intratumoral injection of rat TNF into hamsters bearing Bomirski Ab amelanotic melanoma, a fast growing tumor of high metastatic potential, were tested.
  • In vitro assays indicated a direct cytotoxic effect of TNF against Ab melanoma cells, most probably as an outcome of apoptosis.
  • Intratumoral application of TNF also caused some modulation of cytokine response in melanoma-bearing hamsters as evidenced by increased levels of IL-6 in blood serum.
  • CONCLUSIONS: This study established Bomirski Ab melanoma as a useful model for complex analysis of the anti-tumor activity of TNF.

  • [Title] Anorectal amelanotic melanoma.
  • OBJECTIVE: Anorectal melanoma is a rare, highly malignant tumour with a poor 5 year survival of 10%.
  • Most anorectal melanomas have gross and/or histologic pigmentation, however about 30% of anorectal melanomas are amelanotic.
  • METHOD: We report three cases of amelanotic anorectal melanomas and integrate our data with six case reports of amelanotic malignant melanoma from the literature.
  • Further we compare clinicopathological data and clinical outcome with large series of anorectal melanomas (both, amelanotic and pigmentated).
  • Rectal bleeding was the leading symptom in all cases with a mean duration of 4 months before diagnosis.
  • CONCLUSION: Early diagnosis is key for efficient treatment and improved survival rate for patients with this unusual variant of melanoma.
  • There is no difference in terms of age, time of diagnosis, stage and survival between pigmented and amelanotic anorectal melanoma.

  • BACKGROUND: Anorectal melanoma is a rare and aggressive mucosal cancer.
  • There is usually a delay in diagnosis because about 30% of these cancers are amelanotic and are often mistaken for benign conditions.
  • Herein, we report a case of amelanotic anorectal malignant melanoma with an unusual metastatic deposit in the vulva and also review the literature.
  • Clinical examination showed a pedunculated and ulcerated amelanotic tumour associated with three other nodules, 1 cm in diameter, localized in the vulval mucosa.
  • Histological examination and immunohistochemical staining of all tumours demonstrated malignant melanoma.
  • DISCUSSION: Nine cases of amelanotic malignant melanoma have been reported in the literature.
  • The age at diagnosis ranged from 45 to 77 years.
  • Anorectal melanoma is most common in the rectum, followed by the anal canal.
  • Our case is the tenth case of amelanotic anorectal melanoma and probably corresponds to multiple synchronous primary melanomas of the anorectal region and the vulva, with the possibility that one of the lesions is a primary melanoma and the others are satellite lesions.

  • [Title] Spontaneous apoptosis of melanotic and amelanotic melanoma cells in different phases of cell cycle: relation to tumor growth.
  • Since the spontaneous alteration of native melanotic (Ma) into amelanotic (Ab) transplantable melanoma line it has been observed that this alteration is accompanied by the acceleration of growth of Ab line.
  • Apoptosis estimated by the TUNEL method, alterations in the plasma membrane structure (annexin V staining), changes in the mitochondrial transmembrane potential--delta psi m (JC-1 staining) showed that amelanotic melanoma cells have decreased ability to undergo spontaneous apoptosis.
  • The obtained results showing that in the native melanotic line about 30% of cells are in S+G2/M phases and that 33% of these cells undergo apoptosis could lead to the conclusion that the slower growth of this melanoma line is the result of lower proliferation activity and higher rate of apoptosis of these tumor cells.
  • The number of cells in S+G2/M phases in amelanotic melanoma line increases up to 40% and only 7% of them undergo apoptosis.
  • This observation seems to suggest that the expansive growth of this melanoma line depends mainly on the decreased ability to undergo spontaneous apoptosis, especially in case of cells from S+G2/M phases.
  • Moreover, the obtained results indicate that alteration of melanotic line into amelanotic one, accompanied by differences in many biological features also concerns basic cell processes such as cell cycle and cell death.

  • The compounds prepared were evaluated for their inhibition of NO production, antioxidant activity and also for their ability to inhibit in vitro the growth of four human tumor cell lines: large lung carcinoma (COR-L23), alveolar basal epithelial carcinoma (A549), amelanotic melanoma (C32) and melanoma (A375).
  • Compound 5a was the most active against melanotic melanoma (IC(50) = 11.8 microM) while the other compounds exhibited weak cytotoxicity with IC(50) values >50 microM on all cell lines.

  • BACKGROUND: Malignant melanomas are intrinsically resistant to many conventional treatments, such as radiation and chemotherapy, for reasons that are poorly understood.
  • RESULTS: Endogenous melanogenic cytotoxicity, produced by damaged melanosomes, resulted in pronounced cell growth inhibition in MNT-1 cells compared with amelanotic SK-MEL-28 cells.
  • CONCLUSIONS: Melanosome dynamics (including their biogenesis, density, status, and structural integrity) regulate the drug resistance of melanoma cells.
  • Manipulation of melanosome functions may be an effective way to enhance the therapeutic activity of anticancer drugs against melanoma.

  • alpha-Melanocyte-stimulating hormone (alpha-MSH) is known to bind to the melanocortin receptor 1 (MC1R) which is overexpressed on melanotic and amelanotic melanoma cells. alpha-MSH analogues are potential candidates for specific targeting of melanoma metastases.
  • The peptides were evaluated in vitro for MC1R binding and bioactivity and, after labeling with (111)In, for in vitro cellular uptake and in vivo tissue distribution in mice carrying B16F1 melanoma tumors.
  • The glycopeptides showed excellent binding affinities in the low nanomolar to subnanomolar range using both murine and human melanoma cell lines.
