  • BACKGROUND: In a long-term retrospective immunohistochemical study of adenoid cystic carcinoma (ACC) of salivary gland, we investigated the relation of p63 immunodetection to prognosis.
  • CONCLUSIONS: To our knowledge, the present study is the first to report prognostic significance of p63 in salivary gland ACC and the first report of a robust and well-studied immunohistochemical stain performable on routinely fixed and processed tissue with prognostic utility.

  • SMAD4, also known as DPC4 (deleted in pancreatic carcinoma 4) was initially identified as a tumor-suppressor gene.
  • It is located on 18q21, a region frequently deleted in pancreatic carcinoma.
  • Functionally inactivating mutation of the gene also occurs in many cases of pancreatic cancer.
  • Functional loss of SMAD4 is frequently detected not only in pancreatic carcinoma but also in colorectal carcinoma.
  • The purpose of this study was to elucidate the role of SMAD4 in adenoid cystic carcinoma of the salivary glands.
  • We examined 34 cases of adenoid cystic carcinoma for loss of heterozygosity (LOH) of the SMAD4 locus and for the existence of mutations of the gene.
  • In conclusion, the infrequent LOH of the SMAD4 gene locus and lack of SMAD4 mutations indicate that SMAD4 does not play a role in the tumorigenesis of adenoid cystic carcinoma of the salivary gland.

  • Mucoepidermoid carcinoma (MEC) and adenoid cystic carcinoma (ACC) are salivary gland neoplasms with divergent morphological features and clinical behavior.
  • In previous studies, we have shown concordant loss of Fhit and Wwox expression in breast cancer, with significantly more frequent loss in cancers of basal-like phenotype.
  • To determine if there is a similar association in salivary gland neoplasms, we designed a study of MEC and ACC of salivary gland on tissue microarrays (TMA).
  • TMAs were constructed from 25 MEC and 19 ACC of salivary gland.
  • The results suggest that, as for breast cancer, loss of Fhit and Wwox expression might have a role in the pathogenesis of basaloid differentiation in salivary gland neoplasms; alternatively, differences in chromatin structure at chromosome fragile regions might make fragile genes more accessible to DNA damage and rearrangement early during preneoplastic stages of basaloid cancers.

  • CONTEXT AND OBJECTIVE: Salivary gland tumor management requires long-term follow-up because of tumor indolence and possible late recurrence and distant metastasis.
  • Adenoid cystic carcinoma (ACC) accounts for 10-15% of such tumors.
  • DESIGN AND SETTING: Retrospective study at Head and Neck Service, Universidade Estadual de Campinas.
  • Major salivary gland tumor extent was routinely assessed by preoperative ultrasonography.
  • Diagnosis, surgery type, margin type (negative/positive), postoperative radiotherapy and recurrence (presence/absence) were evaluated.
  • RESULTS: There were eleven major salivary gland tumors (52.3%), seven submandibular and four parotid.
  • Ten patients (47.7%) had minor salivary gland ACC (all in palate), while the submandibular was the most frequently affected major one.
  • CONCLUSION: Adenoid cystic carcinoma has locally aggressive behavior.

  • Adenoid cystic carcinoma (ACC) is an uncommon tumor of the head and neck that may occur in any salivary gland tissue.
  • ACC mostly occurring in the major and minor salivary glands, has some unique characteristics such as slow growth, diffuse invasion, and high incidence of distant metastasis.
  • It is a high malignant carcinoma characterized by intensive local invasion and insidious distant metastasis to the lung at an early stage, which is responsible for a poor long-term survival rate.
  • The main clinical treatment to adenoid cystic carcinoma depended on surgical operation in the past.
  • However, it was not so easy to completely excise adenoid cystic carcinoma which resulting in the residual of tumor cells.
  • Adenoid cystic carcinoma is not sensitive to conventional chemotherapeutics, so it is necessary to explore a new kind of drug which possesses inhibition and killing effects to this tumor.
  • It also induces apoptosis in a variety of cancer cells over a wide dose range.

  • OBJECTIVE: To examine the expression of Ezrin in human salivary gland adenoid cystic carcinoma and investigate the effects of Ezrin gene silence on cell proliferation, apoptosis and invasion of adenoid cystic carcinoma (ACC)-M.
  • METHODS: The expression of Ezrin was detected by immunohistochemistry in normal salivary gland tissue (n=15), pleomorphic adenoma (n=40) and salivary gland adenoid cystic carcinoma (n=43).
  • RESULTS: The positive rate of Ezrin expression in ACC was significantly higher than that in normal salivary gland tissue and pleomorphic adenoma (P<0.05).
  • CONCLUSIONS: Over expression of Ezrin in human salivary gland adenoid cystic carcinoma may promote genesis, development and metastasis of tumors.

  • The CD117 (KIT) protein is overexpressed in many human neoplasms including adenoid cystic carcinoma of salivary glands.
  • To evaluate the function of c-kit-activating mutations in adenoid cystic carcinoma of the salivary gland, we studied 14 cases (13 primary, 1 cervical lymph node metastasis) from our institution.
  • This study is the first to report c-kit gene mutations in primary adenoid cystic carcinoma of the salivary gland.
  • Identification of such potential gain-of-function mutations in exon 11, and less frequently in exons 9, 13 and 17, suggests that KIT may be involved in the pathogenesis of adenoid cystic carcinoma of salivary glands.
  • Our study raises a prospect of correlation of c-kit mutation and a potential treatment of adenoid cystic carcinoma with tyrosine kinase inhibitor (imatinib).

  • [Title] RNAi knockdown of C-erbB2 expression inhibits salivary gland adenoid cystic carcinoma SACC-83 cell growth in vitro.
  • OBJECTIVE: To knockdown the C-erbB2 gene in salivary gland adenoid cystic carcinoma SACC-83 cells using RNA interference, and determine the effect of silencing C-erbB2 on cell proliferation.
  • RESULTS: Compared with the control, C-erbB2 mRNA expression was decreased in the C-erbB2-siRNA transfection group, and immunohistochemical analysis indicated that C-erbB2 protein expression was decreased.
  • The differences were statistically significant (P < 0.05) between the C-erbB2-siRNA group and the control groups.
  • Following the C-erbB2 knockdown, the percentage of apoptotic cells was 5.63% compared with 2.04%, 2.85%, and 2.98% in the three control groups, respectively.
  • CONCLUSION: RNA interference can effectively silence C-erbB2 gene expression and inhibit growth of SACC-83 cells, which indicates the potential of targeting this gene as a novel gene therapy approach for the treatment of salivary gland adenoid cystic carcinoma.

  • [Title] [Evolution pattern of the Runx3 gene 5'-CpG island methylation in human salivary gland adenoid cystic carcinoma].
  • OBJECTIVE: To investigate the evolution pattern of the Runx3 gene 5'-CpG island ~3478 bp region methylation in human salivary gland adenoid cystic carcinoma (SGACC).
  • METHODS: Quantitative MSP method was used to detect the methylation status of CpG island in various regions (No.1-10) of Runx3 promoter region, and Western blot was used for detection of the expression of Runx3 protein in 41 salivary gland SGACC samples and corresponding non-neoplastic salivary gland tissues.
  • A Logistic model was used to analyze the risk ratio between the methylation status of CpG island in Runx3 gene and development of salivary SGACC, meanwhile, the possible association among the methylation of Runx3 gene, the clinicopathological parameters of SGACCs, and Runx3 protein expression was compared.
  • The methylation rate was highest in region No. 1 and No. 2 among the successive ten regions ranging from the 5' region to the transcription start site within the Runx3 CpG island, and lowest in the transcription start site both in SGACCs and normal salivary glands.
  • Furthermore, there was no methylation in the transcription start site in nomal salivary glands tissues.
  • Western blot results revealed that the Runx3 protein level in SGACC was significantly lower than that in normal salivary glands (P < 0.01).
  • CONCLUSIONS: Methylation of the Runx3 CpG island spreads from the most 5'-region to the transcription start site in human salivary gland adenoid cystic carcinoma, and the transcription start site may be a critical region for the methylation of Runx3.

  • BACKGROUND: The role of promoter methylation in the inactivation of E-cadherin (E-cad) in salivary gland adenoid cystic carcinoma (ACC) is unknown.
  • The objective of this study was to determine the role and potential clinical implications of promoter methylation of E-cad in salivary gland ACC.
  • METHODS: The promoter methylation status of E-cad was determined by using methylation-specific polymerase chain reaction (PCR) analysis in 60 primary salivary gland ACC tissues and 3 ACC cell lines.
  • CONCLUSIONS: E-cad frequently is inactivated in salivary gland ACC through promoter methylation, and E-cad promoter methylation may play a role in tumor cell differentiation and perineural invasion.

  • OBJECTIVE: To investigate the differently expressed Homeobox genes between adenoid cystic carcinoma of salivary gland and normal gland tissue, and find out the effect of homeobox genes on oncogenesis and differentiation of adenoid cystic carcinoma of salivary gland.
  • METHODS: Six strictly paired specimens including adenoid cystic carcinoma and its surrounding normal gland tissue and two pairs of specimens including cell strain of adenoid cystic carcinoma and its surrounding normal gland tissue were established.
  • RT-PCR technique was used to examine the mRNA expressing level of highly suspected relevant genes of adenoid cystic carcinoma in different specimens.
  • RT-PCI showed that there was statistical expressing difference between TGIF, EVX1 and normal gland tissue in ACC-M.
  • CONCLUSION: As the key gene to cellular proliferation and differentiation, homeobox genes are closely relevant to the oncogenesis of adenoid cystic carcinoma of salivary gland.

  • Adenoid cystic carcinoma is a tumor of exocrine glands originating primarily from the minor and major salivary glands, lacrimal gland, bronchus, breast, and intestinal and genital tracts.
  • Intracranial remote metastasis from adenoid cystic carcinoma in salivary gland is quite rare.
  • The authors encountered a case of intrasellar remote metastasis from an adenoid cystic carcinoma of parotid gland origin, presenting with hyponatremia secondary to the syndrome of inappropriate secretion of antidiuretic hormone (SIADH).
  • To our knowledge, this is the first reported case of metastasis from an adenoid cystic carcinoma to intrasellar area.
  • A 78-year-old woman had an adenoid cystic carcinoma in the left parotid gland, which was resected surgically followed by local radiation therapy of 60 Gy.
  • Histopathological examination of the tumor specimens revealed adenoid cystic carcinoma, which had identical histological findings as those of the painful superficial cervical lymph nodes resected in the same operation.
  • Although rare, metastasis from tumors including those of salivary gland origin should be considered in the differential diagnosis of unusual pituitary tumors.

  • Tumors with cribriform appearance, similar to that of salivary gland adenoid cystic carcinoma, have been described at various anatomic sites.
  • The mass displayed a prominent cribriform architecture, akin to adenoid cystic carcinoma with an immunophenotype that supported a urothelial origin.
  • This cribriform urothelial neoplasm of the renal pelvis may, in fact, represent a variant of an inverted urothelial neoplasm with a prominent cystic component or florid ureteritis cystica.

  • BACKGROUND: Fine-needle aspiration biopsy (FNAB) is useful in diagnosis of major salivary gland adenoid cystic carcinoma (ACC), but it has been rarely reported on minor salivary gland ACC.
  • OBJECTIVE: The aim of this study was to report the utility of FNAB on diagnosis of ACC involving minor salivary glands.
  • Diagnosis of the 3 cases was subsequently confirmed through histopathology.
  • CONCLUSION: The FNAB was easy to perform and cost-effective, and proved to be an useful adjunctive tool in diagnosis of intraoral ACC.

  • A choroidal biopsy was consistent with metastatic adenoid cystic carcinoma from her submandibular salivary gland treated 5 years previously.
  • All of the metastases demonstrated a basaloid histologic variant of adenoid cystic carcinoma that is known for its aggressive potential.
  • Salivary gland carcinoma rarely metastasizes to the choroid; however, evaluation of patients with this carcinoma should include ophthalmic examination.

  • PURPOSE: To investigate the expression of focal adhesion kinase(FAK) and PTEN in salivary adenoid cystic carcinoma (SACC) and its significance.
  • RESULTS: The expressions of FAK in salivary adenoid cystic carcinoma was 94%, and the negative expressions of PTEN was 70%.There was a significant difference between normal salivary gland and salivary adenoid cystic carcinoma (P<0.01).

  • The aim of the study was to analyze Maspin expression in salivary gland cancer as well as its prognostic impact on survival in comparison to clinical parameters.
  • Immunohistochemical staining was carried out in 73 cases of salivary gland malignancies.
  • High proportions of Maspin expression were observed in adenoid cystic carcinomas, mucoepidermoid carcinomas and carcinomas ex pleomorphic adenoma, low proportions were seen in salivary duct carcinomas.
  • Analysis of the prognostic impact of Maspin expression was restricted to salivary gland carcinoma types of intermediate malignancy grade (adenoid cystic carcinoma, mucoepidermoid carcinoma and carcinoma ex pleomorphic adenoma).
  • According to these findings, Maspin might be useful as a new prognostic marker in adenoid cystic carcinoma and in salivary gland carcinomas with intermediate grade of malignancy where grading systems are still under debate.

  • Esophageal localization of adenoid cystic carcinoma of minor salivary glands is rare; it may occur in the early to mid-sixties' age group and is more frequently encountered in men than women.
  • In the majority of cases, it arises from subepithelial glands of the middle to lower third of the esophagus, a similar distribution as squamous cell carcinoma.
  • We report a case of a patient with metastatic primary esophageal adenoid cystic carcinoma progressing on platinum- and irinotecan-based regimens, who achieved an objective response with oxaliplatin-based chemotherapy in combination with cetuximab.

  • Carcinoma-associated fibroblasts (CAFs) are a key determinant in malignant progression of cancer and represent an important target for cancer therapies.
  • In this work, we present a microfluidic-based 3D co-culture device to reconstruct an in vitro tumor microenvironment and firstly investigate the effect of CAFs on cancer cell invasion in 3D matrix.
  • Salivary gland adenoid cystic carcinoma (ACC) cells and CAFs embedded in matrix were co-cultured without direct contact on the device.
  • It was observed that CAFs promoted ACC cell invasion in 3D matrix in a spheroid fashion, indicating that CAFs play a critical role in cancer invasion.
  • We further demonstrated the effect of MMP inhibitor as an agent against CAF-promoted cancer invasion.
  • Thus, it provides a suitable platform for elucidating the mechanism of CAF-regulated cancer invasion and discovering anti-invasion drugs in a well defined 3D environment.
